The AT-Closed Vial® ecosystem

The AT-Closed Vial® system enables a fully closed aseptic filling process.

At its core is the AT-Closed Vial® as the primary packaging, combined with the AT-Cap as closure, complemented by suitable filling lines and the single-use system AT-Assembly for sterile liquid transfers.

All components are fully aligned, ensuring a consistent, contamination-controlled, and scalable process.

Process

Closed filling and storage process with AT-Closed Vial®

    Vial provision

    Vials are delivered pre-closed, gamma-sterilized, and ready to fill. Manufacturing and assembly take place under ISO 5 conditions, including comprehensive quality control and CCI verification.

    Aseptic filling

    The vial is punctured with a sterile needle through the stopper. Due to its elastic properties, the stopper reseals immediately. Airborne contamination is prevented as the system remains closed.

    Laser re-sealing

    The stopper is resealed using a one-second laser shot, restoring container closure integrity after puncturing.

  • 4

    Capping

    A sterile snap cap is applied to secure the stopper until product use and protect against tampering.

    Cryogenic storage

    After filling, inspection, and labeling, the vials can be frozen (typically at -80 °C) and, if required, stored in liquid nitrogen (LN₂) before delivery to the clinical site.

    Uncompromising container closure integrity (CCI)

    Validated CCI is ensured, which is critical for cryogenic storage. Robustness is achieved through vial design, polymer selection, ISO 5 assembly, and the closed filling method. The COC body and TPE stopper form a chemical bond, enhancing integrity at low temperatures. Validated methods such as dye ingress testing, CT scans, and laser-based headspace analysis confirm CCI.

    Risk reduction in aseptic filling

    The functionally closed process eliminates airborne contamination risks. Unlike open vial methods, filling occurs through a resealable stopper, ensuring the vial remains closed at all times. This approach reduces product loss and significantly improves aseptic safety during filling and finishing.
